And filling in for Robert Downey, Jr.: Will Ferrell.
The upcoming Dreamworks Animation supervillain 'toon, Oobermind, has three new voices, according to Variety. Ferrell replaces the freshly departed Downey as an evil madman who no longer finds life worth living after his archnemesis is accidentally killed. If Ferrell brings Mugatu to the table (his Zoolander) character, then this could be fine. Hard to see Ferrell underselling it, though.

Tina Fey remains in the cast, playing...Vicki Vale, for lack of a better term. The other two new additions are Brad Pitt as Metro Man, the sworn enemy of Oobermind. And since we already know what happens to him, we need another hero of some kind, which is where Jonah Hill comes into the picture. He's been cast as Titan, a superhero who longs to be a supervillain like Oobermind.
For Dreamworks Animation, 2010 will be a potentially big year, launching three films (including another Shrek flick). This year, all the eggs were in the basket of Monsters vs. Aliens, and even though that made a ton of money, we previously chronicled that the studio doesn't think it did well enough to warrant a sequel. So that really raises the bar for Oobermind and How to Train Your Dragon.
There's no official word on what happened to Downey, but I suspect he's just incredibly busy and is getting bigger paychecks elsewhere.
